Study aim
Comparison of the effect of using or not using of artificial oocyte activation on intra-Cytoplasmic sperm injection outcomes in infertile patients with phospholipase C ζ deficiency
Design
In this study, PLCζ will be used as a diagnostic biomarker in couples subjected to intra-cytoplasmic sperm injection (ICSI). The oocytes obtained from patients who are deficient in this protein, will be randomly divided into two parts following the ICSI method. Assisted oocyte activation (AOA) will be used for one part of oocytes (group treatment, n=110), while the other part will not be received AOA (control group, n=110). Finally, the effect of AOA on fertilization rate, embryo cleavage, and embryo quality will be evaluated in preimplantation embryos microscopically and will be compared between groups.
Settings and conduct
Semen samples from infertile men referred to infertility clinic of Taleghani hospital will be assessed by PLCζ immunofluorescence staining.
Participants/Inclusion and exclusion criteria
By evaluating PLCζ, measured by immunofluorescence staining, semen samples with PLCζ deficiency will be determined and entered to the study.
Intervention groups
1- Group treatment: one part of oocytes that will recieve assisted oocyte activation (AOA). 1- Group control: other part of oocytes that will not recieve AOA.
Main outcome variables
The comparison of fertilization rate, embryo cleavage, and embryo quality in preimplantation stage in infertile patients with PLCζ deficiency in studied groups

Description
Intervention= exposing oocytes in calcium ionophore A23187 following ICSI for 10 min and 10mM (sigma-aldrich). A23187 facilitate passing Ca iones from oocyte' s membrane.
