Protocol summary
The aim of this study is to assess the effect of palm pollen on PRDX1 and PRDX6 antioxidant gene expression in oligospermic men. In this study, oligospermic patient whose diagnosed by an urologist, subdivided in case and control groups. control group patient received only conventional treatment, Whereas, case group treated with conventional treatment and Date Palm Pollent component. 2 to 4 days after the last sexual intercourse, semen from 60 individuals will be collected by masturbation. Inform written consent is given to all participant. Samples will be collected before and after intervention. Inclusion criteria are as follows: normal anatomy and infertility due to oligospermia; Exclusion criteria are as follows: genital infection; individuals with prostatitis; inflammation of the ducts; epididymitis; orchidism and mumps; also Genetic diseases affecting fertility such as (Klinefelter, XX men, Noonan syndrome, chromosome Y microdeletion, mitochondrial DNA mutations). To semen analysis, all samples are aliquoted in RNAs- DNAs free tubes containing 1 ml of sperm freez reagent and storage in -80°C nitrogen tank for Molecular testing and measuring enzyme activity in semen.
In this experiment, 1 g of Magza herbal capsules (containing palm pollen and other compounds), will be given to the case groups 4 times a day for a month (after palm pollen usage molecular testing and gene expression will be done).the randomization is done by permutation.Computer assissted semen analysis will be done according to World Health Organization standards.
The primary outcome of this study is change in count, motility and morphology of sperm and change in PRDX1,PRDX6 antioxidant gene expression

هدف از مطالعه بررسی اثرگرده ی نخل بر بیان ژن های آنتی اکسیدانی PRDX1 و PRDX6 در مردان الیگواسپرمیک می باشد. در این مطالعه، بیماران پس از تشخیص به عنوان افراد الیگواسپرم توسط متخصص اورولوژی در دو گروه مورد و شاهد تقسیم می شوند. بیماران گروه شاهد صرفآ تحت درمان های رایج ناباروری قرار می گیرند درحالیکه گروه مورد علاوه بر درمان های رایج ناباروری، تحت درمان با گرده ی نخل قرار می گیرند.2 تا 4 روز پس از آخرین مقاربت جنسی اقدام به جمع آوری نمونه 60 نفر از شرکت کنندگان به صورت خودتحریکی، می نماییم. به هر دو گروه رضایت نامه و برائت نامه داده خواهد شد. جمع آوری نمونه ها در دو نوبت قبل و بعد از انجام مداخله صورت می گیرد. معیار ورود شامل داشتن آناتومی نرمال و ناباروری به علت اولیگواسپرمی می باشد و از جمله معیارهای خروج از مطالعه می توان به وجود عفونت تناسلی؛ افراد دارای بیماری هایی مانند التهاب پروستات؛ التهاب مجاری اپیدیدیم؛ ارکیدیسم؛ اوریون؛ همچنین داشتن بیماری های ژنتیکی موثر در ناباروری(کلاین فلتر،مردانXX.،سندروم نونان،ریزحذف های کروموزوم Y ،جهش DNA های میتوکندریایی) می باشد. به منظور آنالیز مایع سمن تقسیم بندی های بعدی در لوله های جداگانه با حجم 1میلی لیتر از محلول اسپرم فریز در لوله ی عاری از DNase وRNase ریخته می شود و در تانک نیتروژن در دمای 80- ذخیره می شود تا در آزمایشات مولکولی و اندازه گیری فعالیت آنزیم های سمن بکار گرفته شود. در این آزمایش، 1 گرم از کپسول گیاهی مگزا (حاوی گرده ی نخل و ترکیبات دیگر)4 بار در روز به مدت یک ماه به گروه مورد داده خواهد شد (پس از مصرف گرده نیز آزمایشات مولکولی فوق صورت می گیردو بیان ژن های مورد نظر بررسی می گردد).تصادفی سازی نمونه ها بر اساس روش Permutation انجام می گیرد. آنالیز پلاسمای سمن بر اساس استانداردهای سازمان بهداشت جهانی با استفاده از آنالیز کامپیوتری انجام می گیرد. پیامد اولیه مطالعه، تغییر در تعداد، تحرک و مورفولوژی اسپرم و تغییر در بیان ژن های آنتی اکسیدانی PRDX1,PRDX6 می باشد.

بیماران با معیارهای زیر از مطالعه خارج شدند: سابقه اختلالات ژنتیکی و سیستمیک ناهنجاری دستگاه تولید مثل ضربه بیضه سو مصرف الکل و موادمخدر و داروهای باروری حداقل به مدت شش ماه قبل از مشارکت.
Patients were divided in two groups,30 members in each group according to block randomization method.
Participants entered the study after signing a personal and written consent form based on inclusion criteria.Their cement sample was measured before and after treatment with Date Palm Pollen capsule for 74 days in the case group and also the sample of the control group was measured according to the WHO 2010 analysis analysis guidelines. Also, the concentration of 8-Isoprostane,antioxidant genes was examined.

Intervention of case group: Mogza capsule (1 g): Phoenix dactylifera pollen (0.5 gram); Nigella Satvia seed powder(0.3 gram); Colchicum seed powder(0.1 gram); Alpinia seed powder (0.1 gram) that patients are given four times a day.
In addition to standard treatment by a urologist, the case group received 400 mg / kg of Date palm pollen powder in gelatin capsules for 74 days.

Settings and conduct
This study was conducted during 2015. Date Palm Pollen at a dose of 400 mg/ kg in gelatin capsules was used for 74 consecutive days to treat eligible individuals.Ten fertile men was selected as control group. Semen samples were analyzed before and after Date Palm Pollen treatment in the case group and also semen samples were analyzed in the control group according to WHO guidelines. furthermore, the free 8-isoprostane levels and PRDX1 and PRDX6 antioxidant genes expression were evaluated.
Participants/Inclusion and exclusion criteria
Infertile men aged 20 to 45 years were included in the
study We excluded the case with these criteria from the
research: history of genetic and systemic disorders,
reproductive tract abnormality, testicular trauma, alcohol
and substance abuse, and fertility medications at least
for six months before participation.
Intervention groups
In order to intervene in the case group, in addition to
receiving standard infertility medication by a specialist
physician, they also received 400 mg / kg palm pollen
powder in gelatin capsules for two 74 days.

Normal anatomy of the genital area
Infertility due to oligo asthenospermia
Exclusion criteria:
The case with the following criteria were excluded from the study: history of genetic and systemic disorders.

Participants entered the study after signing a personal and written consent form based on inclusion criteria.Their cement sample was measured before and after treatment with Date Palm Pollen capsule for 74 days in the case group and also the sample of the control group was measured according to the WHO 2010 analysis analysis guidelines. Also, the concentration of 8-Isoprostane,antioxidant genes was examined.

Description
Semen samples were obtained twice from the case group, before and after treatment, and once from the control group through masturbation, masturbation and abstinence for three to five days. We then collected fresh semen samples in sterile plastic containers.

Method of measurement
The samples were exposed to 37 ° C for 30 minutes for liquefaction and semen analysis (fluid volume, pH,viscosity, sperm morphology, concentration and motility) was performed immediately according to the WHO guideline. Sex hormones were also examined using ELISA. In addition, in this clinical trial, a blind and comparative semen analysis was performed by a technician blindly.
